Cell culture unit equipped to carry out experiments involving different models of cell lines of animal origin, including cell lines of human origin. The unit consists of a biosafety level 1 (BSL-1) laboratory, a biosafety level 2 (BSL-2) laboratory and a shared washing and sterilisation area. The BSL-1 laboratory consists of: A TELSTAR laminar-flow cabinet, three Thermo CO2 incubators with an infrared sensor and an 80-litre water jacket, and a Lab-Line CO2 incubator for maintaining cell cultures. An absorbance plate reader. A Hettich Rotina refrigerated centrifuge. A freezer capable of temperatures down to -80°C. A fridge with a temperature range from -4 to 20°C. Two Nuaire class II vertical laminar-flow cabinets. The BSL-2 laboratory consists of: A Nuaire class II laminar-flow cabinet. A Thermo Scientific 3111 Water Jacket CO2 incubator with an 80-litre water jacket. A HERMLE Z306 centrifuge. An Olympus IX71 optical microscope.
Health, cell cultures, primary cultures, cell lines, cell viability, MTT
The unit has class II laminar-flow biosafety cabinets (BSCs) suitable for ensuring the sterility of the work area and protecting the researcher from possible pathogenic agents handled inside it. It is also equipped with incubators that allow controlled temperature, humidity and CO2 concentration conditions to be maintained, a freezer, a refrigerator, an autoclave, a centrifuge, thermostatic baths, etc. The unit¿s technical staff rigorously check the equipment and the general facilities to make sure everything is running properly and they carry out preventative maintenance, for which specific operating protocols have been prepared. The parameters relating to the CO2 pressure, humidity and temperature of the incubators, the laminar flow of the cabinets, the temperature of the refrigerators and freezers, as well as other important indicators, are checked daily.
Maintenance of cell lines (cardiomyocytes, fibroblasts, osteoblasts, neurons, glial cells, mesenchymal stem cells, chondrocytes, retina cells, melanoma cells) and human cell lines (glioblastoma cells), primary cultures of mouse tissues or organs (cerebellum, cortex, hippocampus or medulla cells), supply of cell lines, frozen or in flasks, cell viability testing (MTT, XTT, calcein AM), viability analysis on different surfaces and toxicity evaluation of biomaterials and nanoparticles.
